Coping styles in European sea bass: The link between boldness, stress response and neurogenesis ArchiMer
Alfonso, Sebastien; Sadoul, Bastien; Gesto, Manuel; Joassard, Lucette; Chatain, Beatrice; Geffroy, Benjamin; Bégout, Marie-laure.
Coping styles consist of a coherent set of individual physiological and behavioral differences in stress responses that are consistent across time and context. Such consistent inter-individual differences in behavior have already been shown in European sea bass (Dicentrarchus labrax), but the associated mechanisms are still poorly understood. Here, we combine physiological measurements with individual behavioral responses in order to characterize coping styles in fish. Fish were tagged and placed in a tank for group risk-taking tests (GRT) at 8 months of age to evaluate boldness using the proxy latency of leaving a sheltered area towards an open area. A subsample of these fish were individually challenged 16 months later using an open field test (OFT), in...

Early individual electronic identification of sea bass using RFID microtags: A first example of early phenotyping of sex-related growth ArchiMer
Ferrari, Sebastien; Chatain, Beatrice; Cousin, Xavier; Leguay, Didier; Vergnet, Alain; Vidal, Marie-odile; Vandeputte, Marc; Begout, Marie-laure.
Although individual electronic tagging using PIT (Passive Integrated Transponder) tags is well established, it is mainly used for fish > 60 mm in length. Since electronic tagging is an ideal identification method, we used RFID (Radio Frequency Identification) microtags (6 mm in length, 1 mm in diameter, 10 mg in mass) to characterize individual fish from the early stages of their development and throughout their lifecycles. We used sea bass, Dicentrarchus labrax, (105 day-old fish weighing between 100 and 1100 mg) and studied the effects of intra-coelomic tagging in half the population using different endpoints including survival and tag reading, growth over 6 months and swimming responses. Dead fish were counted daily, biometric data were collected at...

Early life behavioural differences in wild caught and domesticated sea bass (Dicentrarchus labrax) ArchiMer
Benhaim, David; Pean, Samuel; Lucas, Gael; Blanc, Nancy; Chatain, Beatrice; Begout, Marie-laure.
Behavioural studies comparing hatchery and wild-caught fish are useful to improve selection for aquaculture and restocking programmes. We examined swimming behaviour characteristics in wild captured and domesticated sea bass juveniles before and after eliciting a startle response at 8 different ages and always on naive individuals. We specifically investigated whether domestication impacts juvenile sea bass behaviour and whether the first months of captivity induce behavioural modifications in wild juveniles. An apparatus was designed to mimic a predator attack by presenting a sudden visual and mechanical stimuli simultaneously in 8 arenas where single individuals were placed and video recorded. The reactivity response was evaluated and different swimming...

Effect of fasting on self-feeding activity in juvenile sea bass (Dicentrarchus labrax) ArchiMer
Benhaim, David; Begout, Marie-laure; Pean, Samuel; Brisset, Blandine; Leguay, Didier; Chatain, Beatrice.
In various experiments under self-feeding conditions, sea bass groups could be divided into three categories regarding feeder actuation: high, low and zero-triggering fish. In all cases few high-triggering fish were responsible for a high percentage of the feed delivery. A question was raised about the role played by feeding motivation in such high-triggering status acquisition. It was approached by applying a 3-week fasting period in order to induce similar negative specific growth rate (SGR) in two groups of fish of similar mean weight but with either a low or a high coefficient of variation for weight (CVw)(T-low: CVw similar to 11%, 3 tanks of 60 fish each; T-high: CVw similar to 20%, 3 tanks of 60 fish each). These groups were created to test the...

Effect of selection for fasting tolerance on feed intake, growth and feed efficiency in the European sea bass Dicentrarchus labrax ArchiMer
Daule, Sophie; Vandeputte, Marc; Vergnet, Alain; Guinand, Bruno; Grima, Laure; Chatain, Beatrice.
Feed efficiency is a major goal for aquaculture sustainability, and selecting fish to genetically enhance this trait would be highly valuable. However, no selective breeding program specifically targeted to feed efficiency exists for farmed fish, mostly because of the difficulty of measuring individual feed intake. However, a negative phenotypic correlation between feed efficiency and weight loss at fasting has been previously demonstrated in sea bass submitted to feed deprivation (FD). We mated sea bass parents selected for their high (FD+) or low (FD-) weight loss at fasting to produce FD+ and FD- progeny, which were reared in a single tank to avoid common environmental effects. At 8 months of age, 1200 of those fish were submitted to three alternating...

Effect of size grading on sea bass (Dicentrarchus labrax) juvenile self- feeding behaviour, social structure and culture performance ArchiMer
Benhaim, David; Pean, Samuel; Brisset, Blandine; Leguay, Didier; Begout, Marie-laure; Chatain, Beatrice.
This study aims to test the influence of size grading on self-feeding behaviour, social structure (measured by the percentage of triggering acts per individual), growth performances, and blood physiological variables of individually passive integrated transponder (PIT)-tagged sea bass juveniles, using a computerized on-demand feeding system coupled with a PIT tag monitoring device. Three consecutive periods of 27 days each were compared: a first period (P1) before grading (6 tanks of 100 fish; 40.2 +/- 8.9 g) followed by a second period (P2) after grading. The protocol applied aimed to create two groups of fish of similar mean weight but with either a low or a high coefficient of variation of weight (CV(w)) corresponding to an imposed difference in social...

Effects of domestication, selection and stress on the energy balance of Sea bass (Dicentrarchus labrax) in aquaculture ArchiMer
Campeas, Arnaud; Millot, Sandie; Chatain, Beatrice; Begout, Marie-laure; Alunno-bruscia, Marianne.
Reared sea bass (Dicentrarchus labrax) represent 13% of the economic weight of the Mediterranean aquaculture production (FAO, 2005) and is a subject of selection program. However, the traditional goal of selection that prevailed in the last century, i.e. increasing productivity, have to be modulated by new objectives such as better feed management or increasing fish welfare. The aim of our study is to evaluate the influence of domestication and selection, and their interactions with stress, on the energy balance in sea bass (Dicentrarchus labrax). Therefore, we used two experimental datasets and analysed their results through a mathematical model. Four strains of sea bass were studied: i) Wild strain, ii) Domesticated strain (first generation obtained by...

Effects of fasting and re-alimentation on gill and intestinal morphology and indicators of osmoregulatory capacity in genetically selected sea bass (Dicentrarchus labrax) populations with contrasting tolerance to fasting ArchiMer

Fasting and refeeding occur naturally in predators but this is largely ignored when dealing with farmed fish. Therefore, the effects of 3-week fasting and re-alimentation (2.5% of the individual body mass) were investigated using two genetically selected populations (F2 generation) of 250 g juvenile sea bass (Dicentrarchus labrax L.). Blood osmolarity, gill and intestinal morphology and expression of the sodium pump (Na+, K+-ATPase, NKA) were studied on two phenotypes showing different degrees of body mass loss during food deprivation: one group losing body mass rapidly during fasting (F +) and the other one limiting body mass loss during the same period (F-). Blood osmotic pressure significantly decreases due to re-alimentation in both groups, but this...

Enhanced brain expression of genes related to cell proliferation and neural differentiation is associated with cortisol receptor expression in fishes ArchiMer
Sadoul, Bastien; Alfonso, Sebastien; Bessa, E.; Bouchareb, A.; Blondeau-bidet, E.; Clair, P.; Chatain, Beatrice; Begout, Marie-laure; Geffroy, Benjamin.
Stress enhances or inhibits neurogenesis in mammals and some fish species. The link between the two processes is still unclear. Most studies have been performed in very specific stressful or altered environments. Despite the known inter-individual divergence in coping abilities within populations, the relationship between the stress axis and neurogenesis has never been addressed in unstressed individuals. Here we correlate brain expression of the pcna (proliferating cell nuclear antigen) and neurod1 (neurogenic differentiation factor 1) genes, two markers of neurogenesis, with transcripts of cortisol receptors in three fish species living in very distinct environments. Within the three species, individuals with the highest expression of neurogenesis genes...

Estimates of heritability and genotype-environment interactions for body weight in sea bass (Dicentrarchus labrax L.) raised under communal rearing conditions ArchiMer
Saillant, Eric; Dupont Nivet, Mathilde; Haffray, Pierrick; Chatain, Beatrice.
Genetic parameters for somatic growth rate in sea bass were estimated in two experiments where 27 (year-class 1997) to 30 (year-class 1998) families of sea bass (3 dams mated with 9-10 sires according to a full factorial design) were raised mixed in the same tanks starting shortly before hatching (48 h post fertilization). Trials were performed under two temperature (experiment 1, 1997: group high temperature (HT) and group low temperature (LT)) and two density regimes (experiment 2, 1998: group high density (HD) and group low density (LD)) with a subset of the mating design (18 families) being reproduced in the two experiments. All the fish were genotyped at 3-6 microsatellite loci which allowed them to be assigned to the individual breeders used in the...

Estimation et amélioration des performances zootechniques de l'élevage larvaire de Dicentrarchus labrax et de Sparus auratus ArchiMer
Chatain, Beatrice.
By gathering specific measurement criteria and an objective analysis of field methods, the main purpose of this work is to help estimate larval Sea Bass (Dicentrarchus labrax) and Sea Bream {Sparus auratus) stock performance raised under intensive conditions. As such, it is a methodology guide zootechnicians can use, in their daily quantitative and qualitative evaluation stock issues. The first part of this work covers defining the precision of current counting methods and establishing their limitations. The quantification methods analysed, as well as the new proposed methods, relate to the major phases of larval farming, from laying to postlarval stage. Three field methods commonly used in aquaculture to quantify egg population are therefore...

The present study aimed to develop nondestructive methods to accurately follow the growth of fish larvae and post-larvae. European sea bass (Dicentrarchus labrax) aged 71-100 days post fertilization (dpf; 20-419 mg) and Nile tilapia (Oreochromis niloticus) aged 14 dpf (10-25 mg) were used as model species. Several measurements were performed on digital pictures of larvae to model body weight. Using body surface area, contour, length and depth, it was possible to estimate accurately the body weight of young European sea bass and Nile tilapia (r² of 0.98 and 0.95, respectively).

Estimation of genetic parameters of feed conversion ratio based on individual phenotypes and genomic data in sea bass ArchiMer
Besson, Mathieu; Allal, Francois; Chatain, Beatrice; Vergnet, Alain; Clota, Frederic; Ferrari, Sebastien; Vandeputte, Marc.
We estimated genetic parameters of feed conversion ratio (FCR) and daily growth coefficient (DGC) using individual phenotypes measured in aquariums under a restricted feeding regime and genomic information on 2,722 SNP markers. The results showed a moderate heritability of both traits (0.26 for FCR and 0.41 for DGC) as well as close to unity negative genetic correlation between them. Thus, FCR of fish in aquariums could be improved by selecting fish for higher DGC. Nevertheless, these results must be taken with caution until we know whether individual FCR measured in aquariums also reflects the FCR of fish reared in groups.

Evaluation of behavioral changes induced by a first step of domestication or selection for growth in the European sea bass (Dicentrarchus labrax): A self-feeding approach under repeated acute stress ArchiMer
Millot, Sandie; Pean, Samuel; Leguay, Didier; Vergnet, Alain; Chatain, Beatrice; Begout, Marie-laure.
Among other strategies to improve fish welfare in rearing environment, domestication and/or selective breeding was proposed to minimize fish responsiveness to husbandry practices. To verify this hypothesis on a recently domesticated specie, the sea bass, Dicentrarchus labrax, L., an experiment was realized, using four populations differing according to their level of domestication or selection: one population produced from wild parents (Wild), one population produced from parents domesticated for one generation (Domesticated) and two produced from parents selected for growth for one generation (Selected A and Selected B). The experiment was carried out over 91 days with 600 fish (50 fish per tank, 150 fish per population). After a control period, the fish...

Exploration behaviour and flight response toward a stimulus in three sea bass strains (Dicentrarchus labrax L.) ArchiMer
Millot, Sandie; Begout, Marie-laure; Chatain, Beatrice.
Domestication and selection may affect fish behaviour, sometime as soon as at the first generation of domestication. However, knowledge about how both processes impact on fish spatial exploration and swimming activity still is to be improved. The objective of this experiment was (i) to evaluate spatial exploration behaviour and swimming activities of three sea bass strains having different domestication and selection levels and (ii) to analyse their responses to an acute stress. Sea bass exploration and swimming activities were studied before, during and 40 min after a stimulation (standardized fall of an object). The experimental tank was divided in to four zones, and the time spent, the distance travelled in each zone and the swimming complexity were...

First Insight into Exploration and Cognition in Wild Caught and Domesticated Sea Bass (Dicentrarchus labrax) in a Maze ArchiMer
Benhaim, David; Begout, Marie-laure; Lucas, Gael; Chatain, Beatrice.
European sea bass aquaculture is so recent that very little is known on the effects of the early steps of its domestication. Behavioural parameters are sensitive indicators of the domestication process since they are generally impacted as soon as the first generation. The present work compared wild-caught and domesticated sea bass juvenile swimming activity, exploration and ability to learn to discriminate between two 2-D objects associated to a simple spatial task that enabled the tested individual to visually interact with an unfamiliar congener (the reward) located behind a transparent wall at the end of one of the two arms of a maze. Ten fish from each origin were individually tested 3 times in a row during 3 days (9 trials in total). Fish were placed...

First links between self-feeding behaviour and personality traits in European seabass, Dicentrarchus labrax ArchiMer
Ferrari, Sebastien; Benhaim, David; Colchen, Tatiana; Chatain, Beatrice; Begout, Marie-laure.
Most studies carried out with seabass under self-feeding conditions report an intriguing social structure that is built around the device and the food dispenser with three coexisting triggering categories: high-triggering (HT), low-triggering (LT) and zero-triggering (ZT) fish. However, neither sex nor feeding motivation or hierarchy can explain the establishment of this specialization. We characterised the personality of seabass with the commonly used restraint and open field tests and assessed the link between personality traits and individual triggering activity towards the self-feeder apparatus. We found no differences between triggering categories during the restraint test but high triggering fish were characterised as shyer than low- and...
